- Add another 1.5ml of culture to pellet, spin, and remove supe (optional)
- Resuspend in 500 ul sterile dd H2O.
- Spin 10 min, remove supe.

How much does miniprep yield?

Is it okay to vortex plasmid DNA?
Vortexing and pipetting your plasmid — take it easy! Nicked or linear DNA may occur due to mechanical shearing of DNA if preps are vortexed or shaken too vigorously during isolation of the plasmid. So take it very easy; mix gently, don’t vortex and pipette softly and sparingly.
Can you freeze bacteria before miniprep?
I did mini prep on bacterial pellets that were frozen for a whole year and it worked fine.
